研究目的は、マクロファージのNO、およびサイトカイン分泌機能が磁界曝露により変化するか否かを解明することである。刺激物質(LPS、IFN-γ)を添加したマウス由来マクロファージを培養しながら50 Hz、14 mTrmsの回転円磁界に24時間曝露し、分泌されたNO、TNF-α、IL-1β、およびIL-6の量を測定した。実験は再現性の確認のため、3回実施した。その結果、一部の実験条件下では各産生量に統計学的に有意な変化が認められた。しかし、その各産生量の増減の方向は一致せず、いずれも繰り返し実験で変化の再現性はなかったことから、本実験条件でのマクロファージ分泌機能に関して一貫した磁界の影響はないことが示唆された。
概要 (英文)
Macrophage plays important roles in host defense and immune system. Its major functions include secretion of nitric oxide (NO) and cytokines, phagocytosis,and antigen presentation. NO is a mediator of the antimicrobial and tumoricidal activities of macrophage. Tumor necrosis factor-alpha (TNF-α), interleukin-1b (IL-1β), and IL-6 are typical examples of multifunctional cytokines involved in the regulation of the immune response, and inflammation. The production of them from macrophage is induced most efficiently in response to lipopolysaccharide (LPS) and interferon-gamma (IFN-γ). There have been so far few studies investigating the effect of magnetic field (MF) exposures on the macrophage functions. Objective of this study is to determine the effect of 50 Hz, 14 mTrms circularly polarized MF exposures on NO and cytokines production in murine peritoneal macrophage (PEM). The experimental design was as follows: PEMs obtained from specific pathogen free, male BALB/c mice were precultured for 2 hours under 37℃ in 5 % CO2 in DMEM supplemented with 10 % fetal bovine serum and antibiotics using 6-well plates. The media were changed to fresh ones with or without LPS, IFN-γ and/or NG-monomethyl-L-arginine (NMMA), an inhibitor of NO production, shortly before the initiation of 24 hours MF exposure. Nitrite is commonly quantitated by the Griess reaction as a measure of NO generation in activated macrophages. Produced three cytokines, TNF-α, IL-1β, and IL-6 were measured with enzyme-linked immunosorbent assay. Comparison was always performed between exposed group and corresponding sham-exposed group. In each experiment, three replications were performed in a blind fashion. Although statistically significant changes due to MF exposures were observed in NO, TNF-α, IL-1β, and IL-6 productions in experiments, the changes were not always replicable. Because the directions of the changes were not constant, and reproducibility of the changes was not manifest, obtained data suggest that macrophage secretion of NO and cytokines was not altered at MF exposure under the examined conditions.
