無菌化したテッポウユリ鱗茎を貯蔵温度・酸素と培地への養分供給を異にする条件において3日~6ヶ月間暗黒化で貯蔵した後,生鮮重,含水率,植物体構成成分(糖,タンパクなど),および酵素活性(GDH,MDH)を調べて各条件の影響を比較した。さらに,これらの鱗片を1ヶ月間通常の成育条件に移して子球を形成させた後に,同様の項目について調査を行った。その結果,貯蔵後の鱗片からの子球形成は,特に糖やデンプンの消費に対して高い負の相関を示した。また,全ての条件下で貯蔵した鱗片から子球の形成が認められたが,養分供給,低温,酸素非供給の各条件により貯蔵中の鱗片における各成分の消費が抑制され,子球形成態を保つために有効なことが示された。

OPEN-AIR CONDITIONS ARE A PROBLEM IN LONG-TERM STORAGE OF LILIUM SP,BULBS DUE TO DAMAGE BY DEHYDRATION AND ATTACK BY MICROORGANISMS.WHILE GERMPLASM CONSERVATION OF ORDINARY SEED SPECIES IS FEASIBLE USING ULTRA-LOW TEMPERATURE,SPECIES WITH POOR SEED VIABILITY,SUCH AS LILIUM SP,ARE SERIOUSLY CONSTRAINED.FIELD MAINTENANCE IS NOT ONLY EXPENSIVE BUT IS ALSO RISKY BECOMSE SERIOUS LOSSES CAN OCCUR DUE.TISSUE CULTURE OFFERS AN ALTERNATIVE METHOD TO CONSERVING GERMPLASM BECAUSE IT HAS DUAL MERITS OF STORING A RELATIVELY LARGE NUMBER OF PROPAGULES IN A SMALL SPACE UNDER ARTIFICIAL CONDITIONS,AND RAPID MULTIPLICATION OF THE PROGENY IN A SHORT TERM.RECENTLY,MANY WORKS HAVE REPORTED IN VITRO AND NON-FROZEN STORAGE SYSTEMS FOR SEVERAL PLANT SPECIES.THESE REPORTS SHOW ONLY THE EFFECT OF STORAGE CONDITIONS ON THE VIABILITY OF EXPLANTS,BUT NOT THE RELATIONSHIPS BETWEEN THE STORAGE CONDITIONS AND THE CONTENTS OF THE EXPLANTS.THE PRESENT STUDY USED BULB SCALES OF LILIUM SP.AS STORAGE EXPLANTS,IN WHICH PLANT HORMONE IS NOT NECESSARY FOR GROWTH.THE EFFECTS OF STORAGE CONSITIONS(LOW TEMPERATURE,5 DEGREE;O2-FREE ATMOSPHERE,N2:CO2=1.1 AND NUTRIENT SUPPLY)ON FRESH WEIGHT AND CONSUMPTIONOF NUTRITIONAL CONTENTS(WATER,TOTAL AMINO ACID,TOTAL SOLUBLE PROTEIN,TOTAL SUGER,TOTAL STARCH,GLUTAMATE DEHYDROGENASE ACTIVITY AND MALATE DEHYDROGENASE ACTIVITY)WERE EXAMINED FOR 180 DAYS.IN ADDITION,THE VIABILITY OF THE EXPLANT WAS ALSO EXAMINED AFTER TRANSPLANTING STORED SCALES TOREGENERATION CONDITIONS. RESULTS SHOW THAT THE QUANTITY OF THE CONTENTS OF THE BULB SCALE,EXCEPT FRESH WEIGHT AND WATER CONTENT ARE A USEFUL INDEX FOR ESTIMATING THE EFFECT OF STORAGE CONDITIONS AND SUBSEQUENT BULBLET GROWTH.EACH OF THE SUGER AND STARCH CONTENTS AND MALATE DEHYDROGENASE ACTIVITY WERE USEFUL INDEXES.LOW TEMPERATURE,O2-FREE ATMOSPHERE AND NUTRIENT SUPPLY REDUCED CONSUMPTION OF THE NUTRITIONAL CONTENTS IN THE BULB SCALE.IN ADDITION,ALL SCALES STORED UNDER EVERY STORAGE CONDITION REGENERATED A BULBLET.HOWEVER,BULBLET
